Acts as a long-acting, non-sedative antagonist at histaminergic H1-receptors, and antagonizes platelet-activating factor (PAF).

✔️ Indications:

Duvent 10mg Tablet is an effective anti-allergic medicine used to temporarily relieve the symptoms of seasonal allergies. 

✔️ Pharmacology

Rupatadine acts as a non-sedative antagonist at histamine H1-receptors, and it has an extended half-life. Additionally, it inhibits platelet-activating factor (PAF). Both histamine and PAF contribute to bronchoconstriction, and increased vascular permeability, and act as mediators in the inflammatory process. Rupatadine's dual mechanism of action provides a more effective therapeutic impact compared to a single antihistamine.

Moreover, Rupatadine inhibits mast cell degranulation triggered by both immunological and non-immune stimuli. It also suppresses the release of cytokines, particularly tumor necrosis factor-alpha (TNF), in human mastocytes and monocytes. These actions contribute to its efficacy in treating allergic conditions and inflammatory responses.

✔️ Dosage & Administration of Duvent 10mg Tablet

• Adult Dose: 10 mg (1 tablet) once daily.
• Child Dose: Children > 12 years: 10 mg (1 tablet) once daily. Not recommended for those below 12 years.

✔️ Interaction

When administered concurrently with ketoconazole or erythromycin, the systemic exposure of Rupatadine increases. Caution is advised when using Rupatadine with these medications and other CYP3A4 isozyme inhibitors. Additionally, caution is recommended when co-administering Rupatadine with statins, CNS depressants, or alcohol. Avoid consuming grapefruit or grapefruit juice simultaneously with Rupatadine.

✔️ Pregnancy & Lactation

• There is no clinical data available regarding the use of Rupatadine during pregnancy.
• Pregnant women should avoid using Rupatadine unless the potential benefits outweigh the potential risks for the unborn child.
• There is no information on whether Rupatadine is excreted in breast milk.
• Therefore, the use of Rupatadine during lactation should be avoided unless the benefits to the mother justify the potential risk to the newborn child.
